View on StockTwits ↗$WVE Sept. 10 call was strategically constructive, even without new clinical data. The key near-term catalyst is FDA’s written feedback on the WVE-006 Phase 3 strategy. Wave is proposing one integrated registrational trial, with an interim analysis potentially supporting accelerated approval while the same study continues toward full approval. Written FDA feedback or meeting minutes are generally provided within approximately 30 days, depending on the meeting format. Favorable alignment could shorten development timelines, reduce capital requirements and reposition WVE-006 as a registrational-stage asset. Its RNA-editing approach could address both the lung and liver manifestations of AATD, with monthly or potentially less-frequent dosing. The market opportunity is substantial. An estimated 30,000–60,000 initially addressable severe, diagnosed patients could represent approximately $4.5B–$15B annually at orphan-disease pricing. Just 5,000 patients treated at $200,000 per year would generate approximately $1B in annual revenue. WVE-007 remains the much larger upside option. Its approximately 160-patient Phase 2a trial targets patients with BMI 35–50 and measures weight, visceral and liver fat, waist circumference, lean mass, HbA1c, lipids and insulin sensitivity. Importantly, management indicated that the WVE-007 GLP-1 combination study is imminent—and this strategy has already been partially de-risked by Arrowhead’s recently published ARO-INHBE data. ARO-INHBE plus tirzepatide produced 9.4% weight loss at 16 weeks versus 4.8% with tirzepatide alone, along with approximately 23% visceral-fat reduction. Although these are not head-to-head data and do not eliminate Wave-specific clinical risk, they provide meaningful human validation that INHBE silencing can enhance incretin-driven weight loss while potentially improving body composition. This strengthens the rationale for combining WVE-007 with a GLP-1 and could make the program strategically attractive to a major obesity company. No transaction has been announced, but positive combination data could support a partnership involving WVE-007, while favorable FDA alignment could separately increase the strategic value of WVE-006. WVE-007 could ultimately address: • Obesity monotherapy • Combination therapy for millions of GLP-1 patients • Post-GLP-1 maintenance with potentially one or two injections annually Management cited approximately 30M Americans with sarcopenic obesity. At only 1% penetration and $4,000–$8,000 annual pricing, that segment could represent approximately $1.2B–$2.4B in revenue; 5% penetration implies $6B–$12B. The post-GLP-1 maintenance opportunity could be even larger. Bottom line: WVE-006 is the nearer-term foundation with multibillion-dollar AATD potential. WVE-007 is the potentially transformative obesity asset, and Arrowhead’s data have partially de-risked the biological and commercial rationale for Wave’s imminent GLP-1 combination study.
