Praxis’s upcoming calcium channel inhibitor drug, Ulixacaltamide, if approved (PDUFA date Jan29, 2027) can be used on a stand-alone basis or as a combo drug with Propranolol to treat essential tremor. When used as a combo therapy, patients achieved a mean change of -1.07 on the modified Activities of Daily Living (mADL11) scale at week 8, compared to -0.61 for patients on propranolol and a placebo. At week 8, 48% of combo therapy patients achieved a clinically meaningful improvement, versus 25% of patients taking propranolol with a placebo. Propranolol is effective in 50% to 60% of patients who try it. About 10% to 15% of people who initially respond well to Propranolol will develop a drug tolerance after a year or more of use, requiring a prescription change.

Relutrigine inhibits persistent sodium current channels. An NDA for the drug is currently being evaluated by the FDA, and is expected to be approved on or near Dec 27, 2026. Relutrigine’s principal competitor is Neurocrine (drug NBI-355), developed by Xenon Pharma. NBI-355 is currently undergoing a ph1b clinical trial. Data from the trial should be presented around the end of 2026.
