Ionis Pharmaceuticals is the leading developer of antisense technology to discover and develop novel drugs. Its broad clinical and preclinical pipeline targets a wide variety of diseases, with an emphasis on cardiovascular, metabolic, neurological, and rare diseases. Ionis and Biogen brought Spinraza to market in 2016 as a treatment for spinal muscular atrophy, and Biogen launched ALS drug Qalsody in 2023. Ionis brought two additional drugs to market via its cardiovascular-focused subsidiary Akcea, including ATTR amyloidosis drug Tegsedi (2018) and cardiology drug Waylivra (Europe, 2019). Ionis and AstraZeneca launched polyneuropathy drug Wainua in 2024. Ionis marked its first two independent launches in 2025 for Tryngolza (for FCS) and Dawnzera (for HAE).

Trading at 16.0× sales vs its 8.1× historical median P/S.
Fair value ≈ $39.77 · price $78.49 today

Notable options flow hitting the tape - mix of high-beta tech + cyclicals + speculative positioning. $DRAM -80C 7/31 exp, $6M flow, 5.01 avg $HD - 380C 9/18 exp, $1.5M flow, 7.6 avg $IONS - 87.5C 9/18 exp, $1.2M flow, 4.26 avg $AOSL - 50C 7/17 exp, $660K flow, 2.58 avg $SPCX - 165C 8/21 exp, $8M flow, 17 avg What stands out here is dispersion: aggressive upside bets in both growth semis and large-cap cyclicals, suggesting traders are still leaning into momentum rather than hedging for downside. Flow like this doesn’t predict direction on its own-but it does show where positioning pressure is building before price confirms it.

View on StockTwits ↗@GMH21 @BigDaddyPickerhat’s an important point. Payer discussions and inclusion of secondary endpoints on-label are essentially building reimbursement logic early. Markets shift from clinical results to payment validation over time. But what matters is actual coverage and adoption, not discussion stage. I’m tracking $IONS payer pathway, follow me if interested, DM for full view.
View on StockTwits ↗@BigDaddyPickerIf you mean $IONS related treatment cost, 3,000/month is a key barrier. Scale depends more on insurance coverage and payer adoption than efficacy alone. Markets often trade reimbursement expectations early. I’m tracking $IONS, follow me if interested, DM for full view.
View on StockTwits ↗$IONS Big catalyst here. Tryngolza just got FDA approval for severe hypertriglyceridemia, expanding from a tiny orphan indication into a ~3M patient market. This is a major step-change: from niche rare disease asset → potential commercial cardiometabolic franchise. Clinical data is strong (up to ~72% triglyceride reduction, lower pancreatitis risk), and importantly, it’s the first approved therapy in this indication with no direct competitor yet. Market reaction (+3%) is still relatively muted compared to the size of the label expansion, which suggests the story may still be early in being repriced. Now it shifts from “pipeline story” to real commercial execution.
