Futibatinib — same structure as pemigatinib, ORR of 42% with median duration of response of 9.7 months,  again single-arm, no OS requirement.
View on StockTwits ↗$CMPX Ivosidenib (Tibsovo) — the closest analog to tovecimig’s exact problem • Approved on PFS as primary endpoint in a trial allowing crossover, with PFS of 2.7 months vs 1.4 months, HR 0.37  — directionally strong PFS, but the OS picture had the exact crossover confound issue you’re dealing with. • Median OS was 10.8 vs 7.5 months with HR 0.79 (p=0.09) — not statistically significant — and this may have been impacted by crossover from placebo to treatment.  The placebo-arm OS got pulled up by crossover, just like COMPANION-002’s 54% crossover rate is suspected of doing. • FDA’s own final efficacy analysis, accounting for crossover, found placebo-arm OS was actually only 5.1 months  — meaning the FDA itself did the crossover-adjustment math and still approved. This is your single best precedent: same tumor type, same confound mechanism, same regulatory division, drug got through.
